Centrosome dysfunction is linked to cancer and neurodevelopmental disorders such as primary microcephaly. Gurkaslar et al. identify CCDC57 as a pleiotropic regulator of centriole duplication, mitosis and ciliogenesis. They show that it mediates its functions by regulating recruitment of centriole duplication proteins to the centrosome and microtubule nucleation.
